I'm not too sure where the previous poster might have found a train or bus for $50 because that doesn't sound reasonable. Assuming you don't want to walk, your options are car, bus, train or plane. All have pros or cons and your question is actually twofold - cheapest and fastest. Driving your own car will cost you more in gasoline than the cheapest mode of public transportation, not to mention the associated cost of wear and tear on your vehicle. You would also require at least two night's accommodation if you drive whereas on public transit you can still cover ground while you sleep. Generally speaking the cheapest method of transit is often the slowest, but not always.
Are you eligible for any discounts? Are you a member of the military? Are you a student? Do you have an ISIC card? Do you have a Hostelling International membership? Are you a senior? All of these can affect costs so you really haven't given us enough information to work with. What dates are you travelling? Is it the high season or low season? Are you booking far enough in advance to qualify for an advance purchase discount? Are you travelling one way only or will you qualify for a discounted return ticket?
If you can get a good airline seat sale it can be cheaper than bus/train as well as saving you at least a day of travel time. Plus, if you spend two days travelling by bus or train you will need to purchase or pack meals along the way which increases your costs. I think you need to give us more information.

Doing a very quick preliminary search I found a flight for $135 ($219 including taxes) that will get you to Winnipeg in 2.5 hours. The cheapest bus ticket I found was $98 ($110 including taxes) and it will take 31.5 hours so you'll have to add in the cost of at least three meals, maybe more. The cheapest train ticket I found was ($193.50 ($218.66 including taxes) and it takes 35 hours so again you will need to purchase at least three meals and there are only three trains per week so the dates may not be convenient.
If it is a while before you travel, keep checking the airline websites daily for price changes and bear in mind that air-travel is usually less expensive mid-week than it is on the weekend.

Airline prices have already gone down. Westjet is currently having a sale and the one-way fare from Toronto to Winnipeg is $129 plus taxes for a total of $202.41.
The cheapest possible way would to walk of course or do a marathon, if your not into that kind of thing, take a train or bus, it cheaper than car because of gas. and its bad for the environment. I have travelled train and bus they are a cheap way to go. hitch king is taking a risk with strangers. you can also carpool. train or bus would be about 50 dollars round trip.What is the cheapest way to travel from Toronto to Winnipeg?
The fastest way is definitely by air with WestJet, 2h30m, but i don't think it's cheap at all.
